![]() I think you took it personally when it was a general observation. Recognizing messed up thinking is great, but the zen family and dogs are better joined through feeling than thinking. Watching the zen characters is no more mental than watching dogs has to be. I'm as silly as the next guy though, having wasted my life contemplating mostly mental stuff. Its the only way for this family to rub off on us by osmosis. But its up to us to be able to tell the difference. Lets not let the academics and churches get away with claiming their interpretations are where its at, thinking they dodged the knife. Time passed, and the stories have come alive now again for a new generation. They were chewing the chewed at that point. Those who "continued" did not continue at all. It had to be left alone, it had become toxic. Once the ways of the zen characters was formulated, it was over. Within a generations of Dahui and Yuanwu laying the stage for koan practice and then pulling out the rug, zen went underground. Developing (through contemplation) conceptual systems is a challenge many enjoy, it can be fun for a while, but its also exhausting, especially in the long run. Never did whole tons of people ever wake up all at once, in fact, the number in the zen cases is in the dozens more than in the thousands.īut the physical gestures and the use of words as pointing devices, damn, I find that so refreshing and really exciting! The world is the reference, no principles necessary. "new educational", well, I would call it something else in the hope that it pointed toward what the zen family was actually doing, but it wasn't "educational" per se, nor was it really all that new, if you look around, but it certainly, in my opinion, was creative in the context of communities of monks. ![]() Maybe it's a creative teachers duty to invent new educational communication channels. When people are outside of the zen family, why do they think they can guess? Why do they want to think their own way of saving all beings is worth hanging on to and evangelizing? Why do they think the zen characters would not throw it away like a dirty rag? There are things the zen characters themselves don't talk about much, to the chagrin of the faithful. If one is going to hide it from others, how much more are they going to hide it from themselves? But it still shows through to those who can see. Add on a dollop of compassion here, a dollop of reasonable there. And then people start managing their image, get crafty. We show ourselves when we don't think we are showing ourselves. In that book, there are not just offensive caricatures of Asian people ("helpers who wear their eyes at a slant") but deeply offensive depictions of African people wearing grass skirts and topknots while barefoot and shirtless. Let's look at "If I Ran the Zoo," another book that is going out of print. In 1978, Geisel altered the image - removed the pigtail and the yellow tint, changed "Chinaman" to "Chinese man" - but I'm not sure it's significantly better. He drew slanty slashes for eyes and tinted the person's skin bright yellow. In the original version of "And to Think That I Saw it on Mulberry Street," he drew a caricature of an Asian person with a pigtail and wooden shoes. But some of them reflect his outdated and offensive views. But there are already millions of copies of these books out there, and if you are determined to find a copy, you can easily do so. The decision to let six of Geisel's books go out of print was made by Dr. The folks in charge of a writer's literary estate make decisions about what is to be done with that writer's work, and their decisions are not always popular - witness the estate of Harper Lee publishing "Go Set a Watchman," a book she firmly had not wanted published. ![]() ![]() ![]() Assuming you have isolated genomic DNA from maize, and you want to digest it with a restriction enzyme that cuts every 4,096 (4,100) bp, how many fragments will you obtain? To answer this question, you need to have an idea of the size of the genome of the plant you are working with. Now, since most restriction enzymes recognize specific sequences of 6 bases long, the probability of finding such a site is (1/4)6 = 1 site in every 4,096 base pairs (bp). This means the probability of detecting an A (adenine) at a particular location is 1/4. However, let us assume that there are 4 bases on any strand of DNA. Thus, one may ask the question, how often does a restriction enzyme cut within a genomic sequence? It is not possible to give an exact answer for this question. More than 500 different restriction enzymes have been identified and can be purchased commercially. (B) Whereas treatment with BamHI produces fragments with “cohesive” or “sticky” ends. (A) Treating the DNA with SmaI results in fragments with blunt ends. Example of how restriction enzymes cut DNA. Although there are several different types of restriction enzymes, those most useful for rDNA technology recognize specific short sequences in DNA and cleave the DNA at that site to produce cohesive (sticky) or blunt-ended fragments (Figure 2). Restriction endonucleases are a group of enzymes derived (primarily) from bacteria. Other compounds, for example, proteins can also be easily separated from DNA by altering the concentration of salt in the extraction buffer.ĭigestion of DNA with restriction endonucleases Therefore, the final step in many DNA purifications protocols involves precipitation using alcohol. However, the polar sugar phosphate groups of the DNA are repelled by non-polar solutions. For example, DNA is negatively charged, making it soluble in aqueous solution. To achieve this, scientists take advantage of the chemical and physical properties of different molecules inside the cell. ![]() These compounds must be separated from the DNA during isolation. Also, tissues and organs from the same plant, or different plants often contain different composition of metabolites, for example, proteins, lipids, and carbohydrates.
